    '최후의 빙하'로 불리는 거대한 빙하가 붕괴되면서 환경 문제, 특히 화석 자원 사용으로 인한 기후 위기를 해결해야 할 필요성이 더욱 분명해졌다. 이로 인해 화학 산업은 목표 제품을 생산하기 위한 지속 가능한 공정을 강조하는 패러다임의 전환을 맞이했다. 이에 따라 재생 가능한 자원을 사용하여 바이오 기반 화학물질, 바이오 연료, 바이오 폴리머를 생산하는 친환경적인 접근 방식인 바이오리파이너리(Biorefinery) 개념이 주목받고 있다. 대사공학을 통해 수많은 미생물을 이용하여 재생 가능한 제품을 생산할 수 있게 되었다. 본 논문은 맞춤형 대사공학 전략을 통해 5-아미노발레르산을 시작으로 글루타르산, 5-하이드록시발레르산, 1,5-펜탄디올로 확장하는 탄소 5개 플랫폼 화학물질을 생산하기 위한 코리네박테리움 글루타미쿰을 이용한 바이오리파이너리 플랫폼 개발을 다룬다.
    1장에서는 미생물 이용 탄소 5개 플랫폼 화학물질 생산에 대한 최근 연구 동향 조사를 통해 라이신 유래 경로가 효율적임을 확인하였다. 또한, 합성 생물학, 시스템 생물학, 진화 공학을 통해 강화된 대사 공학 전략을 통해 탄소 5개 플랫폼 화학물질 생산을 위한 코리네박테리움 글루타미쿰 균주의 능력을 향상시킬 수 있는 방법을 살펴보았다.
    2장에서는 5-아미노발레르산 생산을 위한 코리네박테리움 글루타미쿰 대사 공학 기술을 개발하였다. 효율적인 5-아미노발레스산 생산을 위해 세 가지 5-아미노발레르산 생합성 경로를 평가하였고, 그 중 슈도모나스 푸티다 유래의 라이신 이화경로를 통한 대사회로가 가장 효율적인 5-아미노발레르산 생산 경로임을 확인하였다. 최적의 탄소 공급원으로 포도당이 선택되었고, 반복적인 대사공학적 연구를 통해 유가식 발효에서 73.32g/L의 5-아미노발레르산을 생산할 수 있었다.
    3장에서는 5-아미노발레르산 경로를 글루타르산 생산경로로 확장하였다. 높은 수준의 글루타르산 생합성을 위해 코리네박테리움 글루타미쿰 맞춤형 효율적인 라이신 전환 모듈 및 글루타르산 생산 모듈을 개발하였다. 또한 라이신 생합성을 향상시키기 위한 공학적 인자를 도입하였다. 최종적으로 개발된 재조합 균주는 유가식 발효를 통해 5L 발효조에서 65.6g/L, 파일럿 규모의 유가식 발효를 통해서는 500L 발효조에서 56.0g/L의 글루타르산 수율을 달성하였다. 생산된 글루타르산은 추가로 정제되어 바이오 기반 디옥틸 글루타레이트의 합성에 사용되었다. 후속 평가를 통해 폴리염화비닐의 친환경 가소제로서 디옥틸 글루타레이트의 효과를 확인하였으며, 가죽 코팅에도 적용되어 친환경 가소제로서의 우수성을 확인하였다.
    4장에서는 코리네박테리움 글루타미쿰에서 5-아미노발레르산 생산 대사경로를 5-하이드록시발레르산으로 확장하였다. 이를 위해 다양한 알데히드 환원효소를 평가하고, 부산물 생합성 경로를 억제하는 대사공학 전략을 채택하였다. 최종적으로 개발된 재조합 균주는 유가식 발효에서 52.1 g/L의 5-HV 생산을 달성하였다.

    In face of the alarming decline of formidable glaciers, known as "doomsday glaciers," the imperative to address environmental challenges, particularly the climate crisis fueled by fossil resource use, is evident. This has prompted a paradigm shift in the chemical industry, emphasizing sustainable process for generating target products. The concept of biorefinery has gained momentum, offering an eco-friendly approach to produce bio-based chemicals, biofuels, and biopolymers using renewable resources. Numerous microorganisms, subjected to metabolic engineering, produce essential products from renewable resources. This thesis focuses on developing biorefinery platforms using Corynebacterium glutamicum for C5 platform chemicals, starting with 5-aminovaleric acid (5-AVA) and expanding to glutaric acid (GTA), 5-hydroxyvaleric acid (5-HV), and 1,5-pentanediol (1,5-PDO) through tailored metabolic engineering strategies.
    Chapter 1 conducts a comprehensive review of recent microbial C5 chemical production, identifying L-lysine-derived pathways as efficient. The chapter explores metabolic engineering strategies, fortified by synthetic biology, systems biology, and evolutionary engineering, to enhance C. glutamicum capabilities for the production of C5 chemicals.
    Chapter 2 demonstrates metabolic engineering in C. glutamicum for 5-AVA production. Strategies include evaluating three 5-AVA biosynthesis pathways, with a focus on the Pseudomonas putida davBA pathway. Glucose is chosen as the optimal carbon source, and iterative engineering of the engineered C. glutamicum strains achieves 73.32 g/L of 5-AVA in fed-batch fermentation.
    Chapter 3 extends the 5-AVA pathway to GTA in C. glutamicum by constructing two conversion modules: the L-lysine conversion module and the GTA production module, aiming for enhanced GTA biosynthesis. The study also places emphasis on augmenting L-lysine biosynthesis. The resulting engineered strain achieves 65.6 g/L GTA in a 5 L fermentor through fed-batch fermentation and 56.0 g/L in a 500 L fermentor via pilot-scale fed-batch fermentation. The obtained GTA is subsequently purified and utilized in the synthesis of bio-based dioctyl glutarate (DOG). Further evaluations demonstrate the effectiveness of DOG as a sustainable plasticizer in the polyvinyl chloride plasticization process and in coating genuine leather.
    Chapter 4 demonstrates expanding the 5-AVA pathway to 5-HV in C. glutamicum, evaluating various aldehyde reductases and suppressing by-product biosynthesis pathways. The final engineered strain achieves 52.1 g/L of 5-HV in fed-batch fermentation.
    Chapter 5 delves into metabolic engineering strategies for achieving selective 5-HV production. A synergistic approach, constructing a by-product recycling pathway and integrating a L-lysine conversion module, yields 48.5 g/L of 5-HV. The subsequent conversion of 5-HV into δ-valerolactone demonstrates the practical applicability of the selective 5-HV biosynthesis system in downstream processes.
    Chapter 6 broadens the metabolic repertoire for the production of 1,5-PDO through the intermediates 5-AVA and 5-HV. Through iterative engineering of C. glutamicum, the final strain attains 22.01 g/L of 1,5-PDO production in fed-batch fermentation.
