IMMUNE-MODULATORY EFFECTS OF HUMAN PLACENTA-DERIVED MESENCHYMAL STEM CELLS ON ACUTE BRAIN INJURY

Human placenta-derived mesenchymal stem cells (hpMSCs) regulate immune responses, and this property can be exploited as cellular sources for cell therapies to treat large spectra of neurologic diseases from acute disorders, including acute stage of stroke and traumatic brain injury (TBI), to chronic aging-related diseases. In neurologic diseases, brain cells are exposed to harsh environment including inflammation and hypoxic conditions. Therefore, investigating how hpMSCs have therapeutic effects in this condition is an important issue. Firstly, we investigated the expression profile of hpMSCs cultured under hypoxic conditions and revealed interesting expression changes in various genes involved in immune regulation. Among them, CD200, a positive regulator of anti-inflammatory TGF-β, was highly expressed under hypoxic conditions. We next investigated whether the CD200-positive hpMSCs influence immunosuppressive effects in vitro. The LPS-stimulated hpMSCs dramatically increased CD200 expression. Immune-modulation factors, including TGF-β, IDO, and IL-10, also exhibited increased expression, thus suggesting that CD200-positive hpMSCs play an important role in regulating inflammatory conditions. To determine whether the immune-modulatory function of hpMSCs is driven by CD200, we used siRNA interference experiments. The siCD200 efficiently silenced CD200 expression in hpMSCs, and this inhibition was maintained after LPS stimulation. In contrast, TGF-β, IDO, and IL-10 expression dramatically decreased after CD200 inhibition and exhibited minimal recovery after LPS stimulation. These results indicated that CD200 may be a key factor for immune-modulation functions in hpMSCs. To confirm the effects of hpMSCs in LPS-stimulated microglial BV2 cells, the cells were primed with LPS and then co-cultured with hpMSCs. CD200-positive hpMSCs exhibited inhibition of pro-inflammatory cytokine expression and NO production in co-cultures with LPS-primed BV2 microglia, and this effect was decreased in CD200-silenced hpMSCs. It suggested that hpMSCs suppress microglial activation and associated NO production through CD200. Therefore, we hypothesized that hpMSCs have strong potentials for ameliorating immune related acute damages such as ischemic stroke by modulating immune responses. We transplanted the CD200-positive hpMSCs into ischemic stroke rat models at 24 h after MCAO and the cells were survived for 1 week after transplantation. To evaluate the immune-modulatory effects of hpMSCs on ischemic stroke brain tissue, we analyzed the mRNA expression of pro-inflammatory and anti-inflammatory factors at 1 day and 1 week after transplantation. Transplantation of hpMSCs showed up-regulation of anti-inflammatory factors including CD200, TGF-b and down-regulation of pro-inflammatory cytokines/ chemokines including IL-1b and TNF-a. These findings suggested that the injected hpMSCs gradually improved the immune-modulatory effects of from 1 day to 1 week in stroke. We determined whether the immune-modulatory effects of hpMSCs might be involved in CD200 expression after transplantation in vivo by using immunohistochemistry analysis. CD200 expression was significantly higher in the region transplanted with hpMSCs than controls at 1 week after transplantation. To determine the effects of cell transplantation on inflammatory responses, we assessed microglia and astrocyte activation in an acute model of stroke by using Iba-1 and GFAP staining. Iba-1 and GFAP-positive cells decreased in the boundary region of hpMSCs-transplanted ischemic brain at 1 week after transplantation. These results demonstrated an hpMSC-dependent decrease of Iba-1 and GFAP in the early phase of stroke, and these effects subsequently contributed to neuroprotection. Altogether, these results demonstrated that hpMSCs are capable of restraining both microglial and astrocytic activation in the early phase of stroke and subsequently protecting neurons against MCAO-related damage. Moreover, at 6 weeks after transplantation, decreases in infarct area and improvements in behavior test were confirmed in hpMSCs-transplanted group when compared with sham group. It demonstrated that the immune-modulatory function of hpMSCs in a stroke rat model underlies the therapeutic recovery potential as a cell therapy. 인간 유래 중간엽 줄기세포는 중추 신경계 질환, 혈관 질환, 퇴행성 질환, 종양을 포함한 많은 질환들을 대상으로 치료 및 저하 된 기능을 향상시키는 능력이 알려지면서 각광받고 있다. 최근 연구들에서 줄기세포는 손상 부위로 이동할 수 있으며, 뛰어난 분화능력, 면역 조절 능력, 분비 인자 등을 바탕으로 손상 부위의 비정상적인 세포를 제거 및 대체 할 수 있을 것이라고 보고되고 있다. 세포치료제 개발에 있어 줄기세포는 중요한 역할을 하고 있으며, 각종 줄기세포에 대한 연구가 활발히 진행되고 있지만 그에 대한 자세한 메커니즘은 잘 알려지지 않은 상태이다. 이러한 한계점 해결을 위하여, 본 실험에서는 중간엽 줄기세포의 하나인 인간 태반 유래 중간엽 세포 (human placenta-derived Mesenchymal Stem Cell; hpMSC) 를 분리 배양하였고, 병리적 환경에서 인간 태반 유래 줄기세포의 치료 메커니즘을 연구하였다. 인간 태반 줄기 세포는 배아 중배엽에서 유래 된 것이며, 대개 출생 후에 폐기되기 때문에 윤리적 문제없이 많은 양의 세포를 얻을 수 있어 세포치료를 위한 훌륭한 후보군이며, 성체 줄기 세포로서 매우 유용하다. 또한, 태반 유래 줄기세포는 HLA 분자를 갖지 않기 때문에 다른 줄기세포들에 비해 면역원성이 낮아 세포 치료제로서 유용한 소스가 될 수 있다. 최근 보고된 연구 결과에서 인간 태반 유래 줄기 세포는 면역 반응을 조절하여 뇌졸중, 외상성 뇌 손상, 알츠하이머 같은 만성 노화 관련 신경 질환 등의 신경계 질환에서 치료 효능이 보여지고 있다. 하지만 염증 및 저산소 상태를 포함한 가혹한 병리적 환경에서 태반 유래 줄기세포가 어떻게 치료 효과를 갖는지에 대한 메커니즘은 명확하지 않다. 본 실험에서는 중간엽 줄기세포의 하나인 인간 태반 유래 중간엽 줄기세포가 병리적 환경에서 어떤 치료 메커니즘을 갖는지 확인하기 위해 저산소 (hypoxic) 조건에서 배양 된 태반 유래 줄기세포를 전사체 분석 (RNA-sequencing) 을 통해 유전자 프로파일을 조사하고 면역 조절과 관련된 다양한 유전자에서 발현 변화를 밝혀냈다. 그 중 CD200은 저산소 상태에서 고도로 발현되었고, 항염증성 인자 TGF-β의 양성 조절 인자로 알려져 있다. 우리는 CD200 양성 태반 유래 줄기세포가 세포 수준에서 면역 조절 효과에 영향을 미치는지 연구했다. 병리적 환경을 위해 LPS로 염증 환경 유도하였으며, 태반 유래 줄기세포는 CD200 발현이 유의적으로 증가 되었다. 항염증 인자로 잘 알려진 TGF-β, IDO, IL-10 또한 증가 된 발현을 나타내어 CD200 양성 태반 유래 줄기세포가 염증 환경 조절에 중요한 역할을 한다는 것을 확인 하였다. 태반 유래 줄기세포의 면역 조절 기능이 CD200에 의해 유도되는지 여부를 확인하기 위해 siRNA 실험을 하였다. siCD200은 태반 유래 줄기세포에서 CD200 발현을 억제 시켰으며, LPS 자극에서도 발현의 억제가 유지되었다. 또한, 항염증 인자 TGF-β, IDO, IL-10 발현도 CD200 억제 후 감소하였고, LPS로 인한 염증 유도 환경에서도 CD200 양성 태반 유래 줄기세포에 비해 항염증 인자들의 발현이 감소되었다. 이러한 결과는 CD200이 태반 유래 줄기세포의 면역 조절 기능의 핵심 요소 일 수 있음을 보여주었다. 다음으로, 마이크로글리아 세포주 (microglia cell line) BV2 세포에서 태반 유래 줄기세포의 효과를 확인하기 위해, LPS로 염증환경을 유도한 뒤 태반 유래 줄기세포와 함께 배양 하였다. CD200 양성 태반 유래 줄기세포는 염증성 사이토카인 발현 및 NO 생산의 억제를 나타내었고, 이 효과는 CD200을 억제 시킨 태반 유래 줄기세포에서 그 효과는 감소되었다. 이 결과는 태반 유래 줄기세포가 CD200을 통해 마이크로글리아 활성화 및 NO 생산을 억제하는 것을 의미한다. 이러한 효과는 태반 유래 줄기세포가 면역 반응을 조절하여 허혈성 뇌졸중과 같은 염증 관련 급성 손상을 치료 할 수 있는 잠재력을 가지고 있다고 할 수 있다. 우리는 허혈성 뇌졸중 모델에서 태반 유래 줄기세포의 염증 조절을 통한 치료 효과를 확인하기 위해 중뇌동맥협착 (Middle Cerebral Artery Occlusion, MCAO) 방법으로 허혈성 뇌졸중 쥐 모델을 만들었으며, 24 시간 후에 CD200 양성 태반 유래 줄기세포를 이식하였다. 병변 주위에 이식된 세포는 뇌 조직 안에서1 주 동안 세포를 생존하는 것을 확인 하였으며, 허혈성 뇌졸중 뇌 조직에서 태반유래 줄기세포의 면역 조절 효과를 평가하기 위해 이식 후 1 일과 1 주일에 염증 인자 및 항염증 인자의 mRNA 발현을 분석 하였다. 태반 유래 줄기세포의 이식은 CD200, TGF-b등의 항염증 인자의 발현을 높였으며, IL-1b 및 TNF-α를 포함하는 염증성 사이토카인/ 케모카인의 발현은 억제시키는 효과를 보였다. 이러한 결과는 태반 유래 줄기세포가 뇌졸중에서 1 일에서 1 주 동안 면역 조절 효과를 점차적으로 향상 시킨다는 것을 의미한다. 다음으로 태반 유래 줄기세포의 면역 조절 효과를 면역조직화학법을 사용하여 뇌 조직에서 CD200 발현을 확인 했다. CD200 발현은 이식 후 1 주 조직에 대조군보다 태반 유래 줄기세포를 이식 한 부위에서 유의하게 높았다. 염증 반응에 대한 세포 이식의 효과를 확인하기 위해 Iba-1 및 GFAP 염색을 하여 급성 뇌졸중 모델에서의 마이크로글리아 및 성상 세포 활성화를 확인했다. Iba-1과 GFAP 는 태반 유래 줄기세포 이식 후 1 주일에서 손상된 병변 부위에서 활성이 감소하였다. 이러한 결과는 뇌졸중의 초기 단계에서 태반유래 줄기세포가 염증 조절 효과를 통해 Iba-1 및 GFAP의 감소를 보여 주었으며, 이러한 효과는 이후 신경 보호 효과에도 영향을 주었다. 결론적으로, 태반 유래 줄기세포는 뇌졸중의 초기 단계에서 마이크로글리아와 성상 세포의 활성화를 억제 할 수 있고, 이후 허혈성 뇌 손상으로부터 신경세포를 보호 할 수 있음을 입증하였다. 또한, 이식 후 6 주째에 태반 유래 줄기세포 이식 군에서는 대조군과 비교하여 경색 면적의 감소 및 행동 검사의 개선이 확인되었다. 이러한 결과는 뇌 손상에서 태반 유래 줄기세포는 면역 조절 기능을 통한 치료제로서의 가능성을 확인하였으며, 면역 조절에 있어서 CD200은 중요한 인자임을 확인 하였다. 또한, 여러 면역 관련 질환에 있어 태반 유래 줄기세포를 이용한 새로운 치료 기전 및 전략을 제공할 수 있어 세포치료제로서의 가능성과 치료법을 한층 더 넓혀주는 기초가 되는 결과이다. 핵심되는 말: 태반 유래 중간엽 줄기세포, 면역 조절, 염증, 뇌졸중, CD200
Bioconversion process is the process of treating microorganisms or enzymes involved in fermentation, which aims to produce new functionality. We performed a bioconversion process in Sophorae fructus, which results in immune-stimulatory effects. The bionconverted S. fructus (BSFE) increased the expression of immune modulators, phagocytosis activity and the expression of CD80 and MHC class II in RAW264.7 cells. The TLR4-MAPK/NF-κB signaling pathway is involved in the activation of RAW264.7 cells by BSFE. In splenocytes, the BSFE increased the expression of pro-inflammatory cytokines and macrophage activation. After the bioconversion process, the genistin present in S. fructus before the bioconversion process decreased, and genistein was produced. The BSFE also alleviated the expression of immune modulators in lipopolysaccharide-induced activated RAW264.7 cells, which is considered an effect of the produced genistein by bioconversion process. Taken together, this suggests that BSFE is considered to have immune-stimulatory and anti-inflammatory effects when excessive immune responses occur. 생물전환 공정은 발효에 관여하는 미생물이나 효소를 처리하는 과정을 통해 새로운 기능성을 생성하는 것을 목표로 한다. 본 연구에서는 괴각 (Sophorae fructus)에 생물전환 공정을 수행하였으며, 생물전환된 괴각 추출물 (BSFE)은 면역 자극 효과를 나타내었다. BSFE는 면역 조절제의 발현과 식세포 작용의 활성화, CD80 및 MHC class II의 발현은 상향조절하였으며, 이 반응은 TLR4-MAPK/NF-κB 신호전달 경로의 관여를 통해 발생한다는 사실을 확인하였다. 또한, 비장세포에서 BSFE는 전염증성 사이토카인의 발현과 비장세포 내 대식세포의 활성화를 증가시켰다. 생물전환 과정 이후, 생물전환 과정 이전 괴각에 존재하던 제니스틴 (geinstin)이 감소하고, 제니스테인 (Genistein)이 생성되는 것을 확인하였다. BSFE는또한 lipopolysaccharide로 유도된 RAW264.7 세포의 면역조절제의 과도한 발현을 완화시켰으며, 이는 생물전환 과정에 의해 생성된 제니스테인의 효과로 간주된다. 본 학위논문을 종합한 결과, BSFE는 면역 자극 효과를 띠며, 과도한 면역 반응이 발생할 경우 이를 완화하는 항염증 효과를 가져 면역 작용을 조절하는 것을 확인하였다.
Regulation of CD4+ T cell differentiation by Bifidobacterium species-derived metabolites
Microbiota-derived metabolites play a vital role in mediating the interaction between microbes and host immunity. The Bifidobacterium genus is critical to the regulation of the immune system. Specifically, Bifidobacterium pseudolongum, Bifidobacterium adolescentis, and Bifidobacterium longum subsp. longum have been identified as regulators of CD4+ T cell subsets, including T helper 17 cells (Th17) and regulatory T cells (Treg) involved in adaptive immunity. However, it remains unclear whether their metabolites are involved in immune modulation. Therefore, this study aimed to examine the possibility that the metabolites derived from these three Bifidobacterium species play a role in regulating the differentiation of Treg and Th17 cells. The results of this study demonstrated that the conditioned media of B. pseudolongum promoted the differentiation of Treg cells and upregulated the expression of genes associated with Treg functions. Furthermore, the conditioned media of both B. pseudolongum and B. adolescentis significantly enhanced the differentiation of IL-17A+ Th17 cells. These findings suggest that B. pseudolongum and B. adolescentis may produce immunomodulatory metabolites that regulate the differentiation of CD4+ T cells. 장내미생물에서 유래한 대사물은 미생물과 숙주 면역계 간의 상호작용을 매개하는 데 중요한 역할을 담당한다. 비피도박테리움 속은 장내 세균 중 하나로 면역계 발달과 조절에 중요하다. 특히 Bifidobacterium pseudolongum, Bifidobacterium adolescentis, Bifidobacterium longum subsp. longum은 Th17 세포와 조절 T 세포를 포함하는 적응 면역계의 CD4+ T 세포를 조절한다. 하지만 그들의 대사물이 이러한 면역 조절에 관여할 수 있는가에 대한 연구는 부족하다. 본 연구에서는 이 세가지 비피도박테리움 종의 배양액이 Th17 세포와 조절 T 세포에 분화에 미치는 영향을 조사했다. 그 결과, B. pseudolongum의 배양액이 조절 T 세포 분화를 촉진시키고, 관련된 유전자 발현을 증가시킬 수 있음을 관찰했다. 게다가, B. pseudolongum, B. adolescentis의 배양액이 인터루킨 17 A를 발현하는 Th17 세포의 분화를 향상시켰다. 따라서, B. pseudolongum, B. adolescentis의 대사물이 T 세포에 의한 면역 반응을 조절할 수 있는 가능성을 확인하였다.
Development of exosome surface display technology expressing immune checkpoint modulator

An immune checkpoint blockade is a next-generation anti-cancer drug that attacks cancer cells by targeting immune checkpoint modulators. Immune checkpoint inhibitors exhibit effective anti-tumor activity in some patients, but there are also concomitant side effects. Here, we developed a therapeutic platform technology using exosomes, which has various advantages as therapeutic agents such as target specificity, non-immunogenicity, and multi- functionality depending on the material loaded. This technology suggests that exosomes can regulate the immune system of the tumor microenvironment with ICOSL expressed on the surface through binding to ICOS, which is a receptor expressed on T cells. Collectively, our results suggest the possibility of a new type of cancer immunotherapy, and a combination with immune checkpoint blockade therapy. 면역관문억제제는 면역관문조절인자를 표적으로 하여 암세포를 공격하는 차세대 항 암제이다. 면역관문억제제는 일부 환자에게서 효과적인 항종양 활성을 나타내지만, 항체이기 때문에 수반되는 부작용도 있다. 본 연구에서는 표적 특이적이며 면역원성 이 없고 어떤 물질을 탑재하는지에 따라 다기능을 나타낼 수 있는 등 치료제로서 다양한 이점을 가지고 있는 엑소좀을 활용한 면역관문억제제 플랫폼 기술을 개발하 였다. 이 기술을 통해 T 세포에 발현되는 리셉터인 ICOS와 결합이 가능하도록 엑소 좀 표면에 공동 자극 분자인 ICOSL을 발현시킴으로써 종양미세환경의 면역 체계를 조절할 수 있음을 시사한다. 종합적으로, 본 연구는 엑소좀 표면에 면역관문조절인 자를 발현시킬 수 있는 기술을 개발하여 기존 면역관문억제제와의 병용투여 뿐만 아니라 새로운 형태의 면역관문억제제로서 활용 가능성을 제시한다.
상단락 방지 기능을 내장한 Converter-Inverter-Brake(CIB) 모듈
This paper introduces a new 25-pin CIB power module with inherent immunity to shoot-through failures. As a result, the need for dead-time compensation in pulse-width-modulated (PWM) inverters that often requires additional current sensors and fast dedicated logic is also eliminated in many applications that now require it. The 25-pin CIB power module was designed, manufactured and tested in 300W inverter system. The experimental results of proposed module and conventional module applied in the same inverter system are directly compared in order to verify its performance. The phase current distortion and THD (Total Harmonic Distortion) are greatly reduced compare to conventional power module. The effects of parasitic impedance due to interconnections in the power module are also investigated using commercially available engineering software, Q3D Extractor and PSPICE. The optimized power module design method are also introduced by investigating the electrical characteristics with a parasitic impedance in a power module such as overshoot voltage, retriggering gate voltage and current ringing. 본 논문에서는 인버터 출력단의 상단락(Shoot-through) 현상을 하드웨어적으로 방지하는 상단락 방지 기능을 내장한 CIB (Converter Inverter Brake) 모듈에 대해 고찰하고, 설계 및 제작하여 실험을 통해 분석하고 특성을 알아보았다. 제안한 모듈은 상단락을 방지하기 위한 dead time이 필요치 않으며 PWM(Pulse Width Modulation) 인버터 동작 시 상단락 현상을 막기 위해 필요한 전류 센싱 보호회로나, 추가적인 보상 알고리즘 등이 요구되지 않는다. 제작한 25-pin CIB 모듈은 300W 인버터 시스템에 적용 평가한 결과, 기존의 시스템에 비해 전류 왜곡 및 총고조파 왜곡이 크게 감소하였다. 또한, 25-pin CIB 모듈 설계에 있어서 상용화 되어 있는 Q3D-Extractor 프로그램을 사용하여 배선간의 기생 임피던스를 추출하고, 이를 이용하여 IGBT 모듈의 스위칭 특성 및 상단락 특성을 시뮬레이션을 통해 알아보았다. 이러한 기생 임피던스의 분석법은 IGBT의 스위칭 동작 시 기생 임피던스에 의한 상단락 전류의 발생여부, 전력소자에 걸리는 최대 전압, Retriggering Margin, 전류 Ringing 현상과 같은 중요 동작 특성을 사전에 분석 및 확인 할 수 있어, 모듈 설계의 최적화 방법을 제시하였다.

패혈증에 대한 미토콘드리아 이식의 효과는 분리된 세포의 유형에 따라 다르다 미토콘드리아 기능은 세포 유형에 따라 다른 특성을 가질 수 있습니다. 이전에는 미토콘드리아 이식이 패혈증 모델에서 면역 조절 효과가 있음을 보여주었는데, 이 연구에서는 패혈증 모델에서 미토콘드리아가 분리된 세포의 유형에 따라 미토콘드리아 이식이 미치는 영향이 어떻게 다른지를 조사하였습니다. L6 (muscle cell), Clone9 (liver cell), UC-MSCs(umbilical cord-derived mesenchymal stem cell) 세 가지 세포로부터 미토콘드리아를 분리하였고, In vitro 및 In vivo 패혈증 모델 을 사용하여 미토콘드리아 이식의 효과를 테스트했습니다. 단핵구 세포주인 THP-1 (monocyte isolated from peripheral blood from an acute monocytic leukemia patient) 세포의 LPS 자극을 In vitro 모델로 사용하였고 첫째, 미토콘드리아 이식 세포에서 미토콘드리아 기능의 변화를 관찰. 둘째, 미토콘드리아 이식의 항염증 효과를 비교. 셋째, 패혈증에서 발생하는 면역저하의 정도를 확인하는 내독소 내성 모델을 이용하여 면역증강 효과를 조사하였습니다. 그리고 In vivo 다균성 패혈증 모델에서 각 세포 유형의 미토콘드리아를 이식하여 생존 및 생화학적 효과를 조사하였습 니다. In vitro LPS 모델에서 미토콘드리아의 산소 소비율을 측정하였을 때 전반적으로 미토콘드리아를 이식한 그룹에서 미토콘드리아 기능이 향상됨을 확인하였고, 세 가지 유형의 세포들 중에서 L6 세포의 미토콘드리아 이식이 미토콘드리아 기능을 크게 향상시켰습니다. 그리고 각 세포 유형에 대한 미토콘드리아 이식은 In vitro LPS 모델의 급성기 (acute phase) 에서 과다염증 (Hyper-inflammation) 을 감소시켰습니다. 또한 내독소 내성 (Endotoxin tolerance) 에서 알 수 있듯이 후기 면역억제 단계에서 면역 기능을 향상시켰습니다. 이러한 기능들은 미토콘드리아 이식에 이용된 세 가지 유형의 세포들 간에 크게 다르지는 않았습니다. 그러나 다균성 복강 내 패혈증 모델에서는 L6 세포의 미토콘드리아 이식만이 대조군에 비해 생존율을 크게 향상시켰습니다. The mitochondrial function could have different characteristics dependent on cell types. Here, we investigated whether the effects of mitochondrial transplantation on the sepsis model could be different depending on the cell type, from which mitochondria were isolated. We isolated mitochondria from L6 muscle cells, Clone9 liver cells and mesenchymal stem cells (MSC). We tested the effects of mitochondrial transplantation using in vitro and in vivo sepsis models. We used the LPS stimulation of THP-1 cell, a monocyte cell line, as an in vitro model. First, we observed changes in mitochondrial function in the mitochondria-transplanted cells. Second, we compared the anti-inflammatory effects of mitochondrial transplantation. Third, we investigated the immune-enhancing effects using the endotoxin tolerance model. In the in vivo polymicrobial fecal slurry sepsis model, we examined the survival and biochemical effects of each type of mitochondrial transplantation. In the in vitro LPS model, mitochondrial transplantation with each cell type improved mitochondrial function, as measured by oxygen consumption. Among the three cell types, L6-mitochondrial transplantation significantly enhanced mitochondrial function. Mitochondrial transplantation with each cell type reduced hyper-inflammation in the acute phase of in vitro LPS model. It also enhanced immune function during the late immune suppression phase, as shown by endotoxin tolerance. These functions were not significantly different between the three cell types of origin for mitochondrial transplantation. However, only L6-mitochondrial transplantation significantly improved survival compared to the control in the polymicrobial intraabdominal sepsis model. The effects of mitochondria transplantation on both in vitro and in vivo sepsis models differed depending on the cell types of origin for mitochondria. L6-mitochondrial transplantation might be more beneficial in the sepsis model. The gut microbiota exerts profound effects on host metabolism and immunity. Interindividual differences in microbial composition, termed enterotypes, shape the metabolic fate of dietary compounds. Red beetroot (Beta vulgaris L.) is rich in betalains, polyphenols, and fermentable fibers with documented antioxidant, anti-inflammatory, and gut-protective properties. However, enterotype-specific biological effects remain insufficiently characterized. This study examined the health-promoting and enterotype-dependent effects of red beetroot using in vitro and in vivo approaches, with emphases on gut microbiota modulation and immune regulation. Red beetroot altered human gut microbial composition and increased short-chain fatty acid production, confirming its role as a fermentable substrate that supports microbial metabolic activity. In mice, red beetroot extract alleviated dextran sulfate sodium–induced colitis by restoring microbial balance, strengthening intestinal barrier integrity, and suppressing NF-κB–mediated inflammatory responses. Enterotype-based analyses demonstrated distinct microbial and metabolic profiles, with Bifidobacterium-rich and lactic acid bacteria–rich communities (Type 1) exhibiting enhanced amino acid metabolism; Prevotella-dominant communities (Type 2) showed more limited alterations. These metabolic differences mirrored immunological effects in macrophages: fermentation supernatants from Type 1 communities enhanced nitric oxide and cytokine production, whereas those from Type 2 communities induced weaker responses. Overall, red beetroot exerts microbiota-dependent and enterotype-specific biological effects, underscoring its potential as a functional food for personalized modulation of gut and immune health. 장내미생물군은 숙주의 대사와 면역에 깊은 영향을 미치며, 개인 간의 장유형(Enterotype)차이는 식이성 화합물의 대사 방식을 결정한다. 레드비트 (Beta vulgaris L.)는 베타레인, 폴리페놀, 그리고 발효 가능한 식이 섬유가 풍부한 식품으로, 항산화·항 염증·장 보호 효과가 알려져 있다. 그러나 레드비트의 장유형 특이적 생리활성 효과는 아직 명확히 규명되지 않았다. 본 연구는 장내미생물 조성과 면역 조절에 초점을 맞추어, 레드비트의 건강 증진 효과와 장유형 의존적 작용을 통합적인 in vitro 및 in vivo 분석을 통해 규명하고자 하였다. 레드비트는 인간 장내미생물 구성을 변화시키고 단쇄지방산(SCFA) 생성을 증가시켜, 미생물 대사활동을 촉진하는 발효성 기질로서의 역할을 확인하였다. 또한 생쥐 모델에서 레드 비트뿌리 추출물은 덱스트란 황산나트륨(DSS)으로 유도된 대장염을 완화시켜, 장내미생물 균형 회복, 장 장벽 기능 강화, NF-κB 매개 염증 반응 억제 효과를 보였다. 장유형 기반 분석 결과, 비피도박테리움과 젖산균이 풍부한 제1형 에서는 아미노산 대사 활성이 증가한 반면, 프레보텔라 우세의 제2형 에서는 변화가 상대적으로 미미하였다. 이러한 대사적 차이는 면역세포 반응에도 반영되어, 제1형 발효 상등액은 대식세포에서 산화질소(NO) 및 염증성 사이토카인 생성을 촉진한 반면, 제2형 상등액은 상대적으로 약한 반응을 보였다. 결과적으로, 본 연구는 레드비트가 장내미생물 구성에 따라 상이한 생리적 반응을 유도하는 장유형 특이적 기능성 식품임을 제시하며, 개인 맞춤형 면역 건강 증진 식이 소재 로서의 잠재력을 강조한다.
