Muscle fatigue is a symptom that decreases your muscles’ ability to perform over time. It can be associated with a state of exhaustion, often following excessive sports activity or exercise. While exercise is a common cause of muscle fatigue, this symptom can be the result of other health conditions. Muscle fatigue caused by excessive activity limits activities of daily living and difficult to maintain a stable posture. Therefore, the purpose of this study was to investigate the effect of stretching, icing, and sports massage on muscle fatigue recovery.
Nutlin-3a Inhibits the Production of CXCL10 by Ionizing Radiation in J774A.1 Murine Macrophages

Nutlin-3a inhibits the interaction between p53, a tumor suppressor protein, and MDM2, an E3 ubiquitin ligase, thereby stabilizing and activating p53. Although the pathophysiological effects of Nutlin-3a, based on its activation of p53, have been documented, studies on its impacts on radiation-exposed macrophages remain sparse. This study explored the modulatory effect of Nutlin-3a on ionizing radiation (IR)-induced CXCL10 production in J774A.1 murine macrophages. Nutlin-3a reduces IR-induced CXCL10 production in these cells and also inhibits IR-induced p38 MAPK activation in the same macrophages. Moreover, using pifithrin-α, a pharmacological inhibitor of p53, it was demonstrated that the inhibitory effect of Nutlin-3a on IRinduced CXCL10 in J774A.1 macrophages is independent of p53. To the best of our knowledge, this study is the first to report the pathophysiological significance that the regulation of p38 MAPK phosphorylation by Nutlin-3a controls IR-induced CXCL10 production in J774A.1 macrophages.

Microwave-assisted Weak Acid Hydrolysis of Proteins

Myoglobin was hydrolyzed by microwave-assisted weak acid hydrolysis with 2% formic acid at $37^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$ for 1 h. The most effective hydrolysis was observed at $100^{\circ}C$. Hydrolysis products were investigated using matrixassisted laser desorption/ionization time-of-flight mass spectrometry. Most cleavages predominantly occurred at the C-termini of aspartyl residues. For comparison, weak acid hydrolysis was also performed in boiling water for 20, 40, 60, and 120 min. A 60- min weak acid hydrolysis in boiling water yielded similar results as a 60-min microwave-assisted weak acid hydrolysis at $100^{\circ}C$. These results strongly suggest that microwave irradiation has no notable enhancement effect on acid hydrolysis of proteins and that temperature is the major factor that determines the effectiveness of weak acid hydrolysis.

γ-Aminobutyric acid (GABA) exists in the brain and spine, increasing acetylcholine, one of the neurotransmitters in the body, and activating blood flow in the brain, leading to an improvement in brain metabolism. The ability of lactic acid bacteria to produce GABA can be judged according to the efficiency of conversion to GABA by Glutamate decarboxylase (GAD) after culturing with the addition of L-glutamate, a substrate, to the medium. In this study, Lactobacillus brevis FBL-3 was used for GABA production. In order to find out the effect of pH, temperature, and substrate addition on GABA production, the seed culture was inoculated into 2.5 L fermentor containing 1 L of medium. GABA was 32.51 g/L when the culture pH was not controlled during fermentation. The optimal conditions for GABA production were found to be pH 5.0 and 37°C, which resulted in 42.52 g/L of GABA. The production of GABA was significantly improved up to 72.121 g/L by adding substrate (10 g/L of glucose and 30 g/L of monosodium glutamate) under the optimized fermentation conditions. The volumetric productivity of GABA was 1.503 g/L/h.

Chitinase 3-like 1 (CHI3L1) has been implicated in the pathogenesis of various diseases, including cancer. In our previous study, we found that anti-CHIL1 antibody inhibited lung tumorigenesis. It has been reported that CHI3L1 is highly overexpressed in colon cancer tissue compared with normal tissue, and high levels of serum CHI3L1 have been associated with worse colon cancer prognosis. We investigated the anticancer effect of an anti-CHI3L1 antibody on colon cancer cells. The anti-CHI3L1 antibody inhibited the cell growth of colon cancer cells in a concentration-dependent manner. The anti-CHI3L1 antibody also reduced the migration but increased apoptotic cell death in colon cancer cells. Using STRING (Search Tool for the Retrieval of Interacting Genes/Proteins), we identified an association between VEGFA and CHI3L1 in colon cancer. We confirmed interaction between VEGFA and CHI3L1 through immunoprecipitation. Furthermore, the combination treatment of the anti-CHI3L1 antibody and VEGFA siRNA inhibited cell growth but increased apoptotic cell death. Additionally, using the Human Base database, we found that CHI3L1 and VEGFA are associated with nicotinamide phosphoribosyltransferase (NAMPT). Furthermore, combining the anti-CHI3L1 antibody and NAMPT siRNA more effectively reduced cell growth and the expression of CHI3L1, VEGFA, and cell growth-related proteins, but significantly increased apoptosis-related proteins. The combination of VEGFA siRNA and NAMPT siRNA more effectively inhibited cell growth. Anti-CHI3L1 antibody inhibited the production of ATP and NADH in colon cancer and had a higher inhibitory effect on these levels when combined with NAMPT siRNA These data demonstrated that anti-CHI3L1 antibody is useful as a potential therapy for colon cancer by inhibiting NAMPT-dependent VEGFA expression and ATP and NADH levels.
