Breviscapine Ameliorates Fear Extinction and Anxiety in BALB/cJ Mice

Objective In this research, the influence of breviscapine on anxiety, fear elimination, and aggression and the potential mechanism was investigated. Methods Anxiety and locomotion were analyzed by elevated plus maze and open field test in mice. Bussey-Saksida Mouse Touch Screen Chambers were used to perform fear conditioning. Territorial aggression was assessed by resident intruder test. Protein levels were evaluated by Western blot. Breviscapine improved fear-extinction learning in BALB/cJ mice. Results Breviscapine at 20-100 mg/kg increased center cross number, total distance traveled, and velocity in a dose-dependent manner. On the other hand, breviscapine at 20-100 mg/kg decreased the immobility time in open field test. In addition, breviscapine at 20-100 mg/kg increased the ratio of time on the open arm, time on the distal parts of the open arm, and total distance traveled in elevated plus maze. Breviscapine at 100 mg/kg increased the average attack latency and decreased the number of attacks over the last 3 days of resident intruder test. In hippocampus, protein levels of postsynaptic density protein-95 and synaptophysin were elevated by breviscapine at these three doses. Conclusion The administration of breviscapine alleviates fear extinction, anxiety, and aggression, while increases locomotor in a dose-dependent manner, which might be associated with its influence on synaptic function.

This study used photochemical reduction to successfully synthesize triangular silver nanoplates (TAgNPs)and silver nanodecahedrons (AgNDs) with higher light-absorption and higher light-scattering properties,respectively, for the same wavelength. To analyze the contribution of light-absorption and lightscatteringof silver nanoparticles to the localized surface plasmon resonance (LSPR) effect, poly(3,4-ethylenedioxythiophene) polystyrene sulfonate was doped with TAgNPs or AgNDs at the same concentration(1.17 lg/cm2) and made into polymer light-emitting diodes (PLEDs). According to the current densityvoltageluminancecharacteristics and electroluminescence (EL) spectra, the enhancement factors for currentefficiency and EL intensity for AgND-containing PLEDs were found to be higher than those for PLEDwith TAgNPs by 24.9% and 138%, respectively. This shows that the metal nanoparticles with higher lightscatteringproperty can induce a relatively strong LSPR effect, which possibly gives a hint to design plasmonicphotovoltaic in future.

Parkinson’s disease (PD) is a neurodegenerative disorder associated with neuroinflammation and gut dysfunction. The G protein-coupled estrogen receptor (GPER) has showed therapeutic potential in inflammatory bowel diseases (IBD), yet its role and underlying mechanisms in PD remain unclear. Here, we aimed to investigate the role and mechanisms of GPER in protecting PD. Female mice underwent bilateral ovariectomies (OVX) and were treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) to induce PD, followed by administration of GPER agonist G1. The expressions of tyrosine hydroxylase (TH) and α-synuclein (α-syn), as well as activations of inflammatory cells and NLRP3 inflammasome in the brain and ileum were evaluated. BV2 cells were pretreated with G1 and/or the antagonist G15, then treated with LPS and ATP to activate NLRP3 inflammasome. Activation of NLRP3 inflammasome in BV2 cells was assessed. Results demonstrated that G1 treatment increased TH expression, reduced α-syn expression, and suppressed inflammation and NLRP3 inflammasome in both the midbrain and ileum of MPTP-treated OVX mice. Pretreatment with G1 suppressed the activation of NLRP3 inflammasome in BV2 cells, while the effect was reversed by G15. These findings indicate that GPER activation exerts a protective effect in MPTP-induced OVX mice by modulating NLRP3 inflammasome in both brain and gut, which might provide novel insights into the pathogenesis and therapy of PD.
Research on the kinematic calibration of the 3-PTT parallel mechanism

Most of the kinematic calibration methods of parallel mechanisms only consider the geometric error of the mechanism, and the calibration effect is poor. To improve the calibration effect, this paper takes the 3-PTT (3-prismatic hook hook) parallel mechanism as the research object and proposes a kinematics calibration method based on the normalized representation model of the geometric error and non-geometric error of the mechanism. First, the degree of freedom of the mechanism is analyzed, its kinematics positive and negative solutions are solved, and its singularity is analyzed. Secondly, a normalized characterization method of mechanism geometric errors and non-geometric errors is proposed, and an error model without redundant parameters is constructed. The end motion error of the machine is measured by laser tracker, and the objective function is constructed. The genetic algorithm is designed to solve the minimum value of the objective function, and the normalized error of the mechanism is identified. By comparing with the recent methods, the better identification performance of the algorithm in this paper is verified. Finally, the kinematics model was corrected according to the identification results. After the calibration was completed, the movement errors of the end of the mechanism along each coordinate axis were reduced by more than 99 %. Compared with other calibration methods, the better calibration performance of the method in this paper is verified.
Protective Immune Response of Bacterially-Derived Recombinant FaeG in Piglets

FaeG is the key factor in the infection process of K88ad enterotoxigenic Escherichia coli(ETEC) fimbrial adhesin. In an attempt to determine the possibility of expressing recombinant FaeG with immunogenicity for a new safe and high-production vaccine in E. coli, we constructed the recombinant strain, BL21 (DE3+K88), which harbors an expression vector with a DNA fragment of faeG, without a signal peptide. Results of 15% SDS-polyacrylamide slab gel analysis showed that FaeG can be stably over-expressed in BL21 (DE3+K88) as inclusion bodies without FaeE. Immunoglobulin G (IgG) and M (IgM) responses in pregnant pigs, with boost injections of the purified recombinant FaeG, were detected 4 weeks later in the sera and colostrum. An in vitro villius-adhesion assay verified that the elicited antibodies in the sera of vaccinated pigs were capable of preventing the adhesion of K88ad ETEC to porcine intestinal receptors. The protective effect on the mortality rates of suckling piglets born to vaccinated mothers was also observed one week after oral challenge with the virulent ETEC strain, C83907 (K88ad, CT+, ST+). The results of this study proved that the adhesin of proteinaceous bacterial fimbriae or pili could be overexpressed in engineered E. coli strains, with protective immune responses to the pathogen.
Transcriptome analysis of tilapia streptococcus agalactiae in response to baicalin

Streptococcus agalactiae (S. agalactiae) is a highly pathogenic bacterial pathogen in aquatic animals. Our previous study has demonstrated the significant inhibitory effect of baicalin on β-hemolytic/cytolytic activity, which is a key virulence factor of S. agalactiae. In this study, we aimed to elucidate the mechanism underlying baicalin's inhibition of S. agalactiae β-hemolytic/cytolytic activity by transcriptomic analysis. Bacteria were exposed to 39.06 µg/mL baicalin for 6 h, and their β-hemolytic/cytolytic activities were assessed using blood plates. Then, the differentially expressed genes (DEGs) were identified and characterized by RNA sequencing (RNA-Seq), and further confirmed using the qRT-PCR. A total of 10 DEGs with 7 significantly up-regulated and 3 significantly down-regulated, were found to be affected significantly under baicalin treatment. These DEGs were associated with 5 biological processes, 5 cellular components, and 3 molecular functions. They were primarily enriched in 3 pathways: lacD and lacC in galactose metabolism, lrgA and lrgB in the two-component system, and ribH/rib4 in riboflavin metabolism. These suggested that baicalin might inhibit the conversion of pyruvate to acetyl-CoA and malonyl-CoA, which are crucial precursors for β-hemolysin/cytolysin synthesis, and result in the accumulation of pyruvate, suppress the expressions of pyruvate cell membrane channel protein genes lrgA and lrgB. Baicalin could compensatory up-regulate the expressions of tryptophan/tyrosine ABC transporter family genes, ABC.X4.A, ABC.X4.P, and ABC.X4.S by inhibiting the expression of cyl A/B in cyl operons. Moreover, it hinders the conversion of D-glucose 1-phosphate to the dTDP-L-rhamnose pathway and leads to a deficiency of L-rhamnose, an important precursor for β-hemolysin/cytolysin synthesis.
Adaptive Sampling Technique for Free-Form Surface of Aero-Engine Blades Based on Discrete Curvature

As the aero-engine blade surface is a typical representative of a free-form surface, its sampling and reconstruction accuracy directly determines the evaluation and correction of subsequent processing technology. In this study, based on a case study of a small engine rotor blade, the blade surface (including concave and convex variations) was divided into thick rectangular meshes based on the idea of mesh subdivision, and the rectangular meshes were refined into primary triangular meshes by Delaunay triangulation. Second, the local Gaussian curvature of the vertices of each primary refined triangle mesh was estimated by combining the Moreton-Sequin method. Then, according to the curvature change in the two connected sampling points on the edge of each primary refined triangle, the adaptive sampling point interpolation along with the curvature change on the boundary of the original triangulation triangle and the densified sampling point distribution, which satisfies the idealized triangulation requirement, were further realized. This approach happens through continuous iteration by introducing a curvature relaxation factor and a greedy algorithm. This method can be used to adaptively adjust the distribution and number of sampling points according to the change in the geometric characteristics of the free-form surface and the curvature distribution of each part. The comparison between this method and the mainstream equidistant sampling method in the blind sampling method and the recent mainstream adaptive sampling method, which uses biquartic B-spline curves, demonstrates that the method proposed in this study can accurately and efficiently represent the complex special-shaped free-form surface in blade free-form surfaces with the fewest sampling points while achieving the highest sampling efficiency and the highest model reconstruction precision.

Most of the gas exchange in the human body is carried out by the lungs, and the physiological activities of the lungs are uninterrupted. Due to the deterioration of the external environment, pulmonary cell lesions are common clinical lung diseases. Mechanical cyclic stretching is one kind of bionic technology to observe lung cancer cells. The A549 cell line is the human lung adenocarcinoma cell line derived from a primary lung tumor. This study investigated the effects of mechanical cyclic stretching on A549 cell activity and gene expression profile. Whereas mechanical cyclic stretching had no significant difference in colony formation and cell migration of A549 cells, the cell invasion increased significantly in A549 cells after stretching. In addition, the microarray data showed that mechanical cyclic stretching altered gene expression, induced inflammation of cells, and activation of Wnt/β- catenin and tumor necrosis factor pathways. More importantly, mechanical cyclic stretching activated the expression of tumor necrosis factor-alpha (TNF-α) protein. Therefore, the increase of cell invasion induced by mechanical cyclic stretching might be associated with the activation of TNF-α in human lung adenocarcinoma cells.
Down-regulation of SENP1 Expression Increases Apoptosis of Burkitt Lymphoma Cells

Objective: To investigate the effect of down-regulation of Sentrin/SUMO-specific protease 1 (SENP1) expression on the apoptosis of human Burkitt lymphoma cells (Daudi cells) and potential mechanisms. Methods: Short hairpin RNA (shRNA) targeting SENP1 was designed and synthesized and then cloned into a lentiviral vector. A lentiviral packaging plasmid was used to transfect Daudi cells (sh-SENP1-Daudi group). Daudi cells without transfection (Daudi group) and Daudi cells transfected with blank plasmid (sh-NC-Daudi group) served as control groups. Flow cytometry was performed to screen GFP positive cells and semiquantitative PCR and Western blot assays were employed to detect the inference efficiency. The morphology of cells was observed under a microscope before and after transfection. Fluorescence quantitative PCR and Western blot assays were conducted to measure the mRNA and protein expression of apoptosis related molecules (caspase-3, 8 and 9). After treatment with $COCl_2$ for 24 h, the mRNA and protein expression of hypoxia inducible factor -$1{\alpha}$ (HIF-$1{\alpha}$) was determined. Results: Sequencing showed the expression vectors of shRNA targeting SENP1 to be successfully constructed. Following screening of GFP positive cells by FCM, semiqualitative PCR showed the interference efficiency was $79.2{\pm}0.026%$. At 48 h after transfection, the Daudi cells became shrunken, had irregular edges and presented apoptotic bodies. Western blot assay revealed increase in expression of caspase-3, 8 and 9 with prolongation of transfection (P<0.05). Following hypoxia treatment, mRNA expression of HIF-$1{\alpha}$ remained unchanged in three groups (P>0.05) but the protein expression of HIF-$1{\alpha}$ markedly increased (P<0.05). However, in the sh-SENP1-Daudi group, the protein expression of HIF-$1{\alpha}$ remained unchanged Conclusion: SENP1-shRNA can efficiently inhibit SENP1 expression in Daudi cells. SENP1 inhibition may promote cell apoptosis. These findings suggest that SENP1 may serve as an important target in the gene therapy of Burkitts lymphoma.
Research on Task Scheduling Convergence Non-Dominated Sorting Method in Cloud Computing

Cloud computing task scheduling is a multi-objective decision on how to perform a variety of tasks simultaneously reasonable sort is essential. Therefore, this article constructs a population of convergence non-dominated sorting method. This method is based on non-dominated sorting method; the use of distributed estimation method is improved by four steps to complete the task order scheduling. The experimental results show that this method has good convergence and distribution.
