Non-invasive Deactivation and Spatial Control of Intracellular Chromosome with Magnetic Nanoparticle
The regulation of cell signaling pathway is important for understanding biological systems. Thus, various techniques to modulate cell signaling by spatial control of cellular components have been developed. Magnetic nanoparticles are emerging as promising candidates in a broad range from cell biology to biomedical application due to their unique characteristics such as remote control and their ability to interact with various cellular and molecular level of interest through their surface decoration. With the advent of advanced nanoscience and nanotechnology in the reliable production and specific tailoring of functional magnetic particles, successful applications were dedicated to a broad range of biomedical applications such as high throughput separation of biomolecules, magnetic resonance image, hyperthermia for cancer therapy, and drug delivery. Recently, fluorescent magnetic nanoparticles inside a living cell were shown to be directly attracted by an external magnetic field. In addition, the signaling transduction pathway was remotely triggered by the functionalized magnetic nanoparticles and the microtubule nucleation and assembly inside Xenopus oocyte was spatiotemporally controlled by magnetic nanoparticles conjugated with key regulatory proteins. Moreover, it is shown that the remote induction of cell death by targeting intracellular lysosome was triggered by functionalized magnetic nanoparticles. In this study, we demonstrate a novel strategy to non-invasively regulate chromosome activity by targeting genetic regulation materials (i.e., oocyte-specific linker histone H1 protein) in live oocyte with the functionalized magnetic nanoparticles and to spatially control the targeted chromosomes by a remote magnetic field. To this end, bacterial magnetic nanoparticles (BMPs, ~50 nm) produced by magnetotactic bacteria (Magnetosome) were used because of their peculiar features, such as high magnetism, high dispersal ability in an aqueous media and good biocompatibility. In addition, BMPs can effectively be conjugated to the diverse biomolecules due to the abundance of amine groups on particles surrounded by lipid membrane. First, we present a novel method to regulate the chromosome activity in mature mouse (Metaphase II) oocytes by targeting the histone H1 protein in chromosome with functionalized magnetic nanoparticles. BMPs were conjugated with histone H1 antibodies (H1-BMPs), specifically targeting the oocyte-specific histone H1 protein. The dose-dependent cytotoxicity test for H1-BMPs was initially conducted before starting experiments. Our data clearly show that the oocytes are able to be tolerated up to the delivery of 1 μg/ml H1-BMPs without any developmental retardation and this concentration is optimal for regulation of a chromosome activities. In the first stage, to verify this approach, we examined whether H1-BMPs have the specific affinity against the chromosomal histone H1 protein in live oocyte. The capability of H1-BMPs for specific targeting was easily detected by confocal microscopy. In addition, we investigated how oocyte developmental process was affected by the magnetic labeling of chromosomes. The parthenogenesis was induced to activate the cell cycle progression. When chromosomes were targeted by H1-BMPs, oocytes were developed up to the 4-cell stage, but began to degrade afterwards and failed to reach the blastocyst stage regardless of the application of magnetic field. Interestingly, in mouse oocyte, it is well known that transcriptional repression in parthenogenesis is proceeded until four cell stage and newly synthesized ribonucleic acid (RNA) begins to be transcriptionally expressed at late four cell stage. Thus, we believed that an abrupt fall in oocyte development after four cell stage is caused by the perturbation of transcriptional activity. Second, we demonstrate the spatial control of targeted chromosomes with H1-BMPs and remaining unbound H1-BMP inside living oocyte via a remote magnetic field. In these experiments, immature oocytes at germinal vesicle breakdown (GVBD) stage were selected for subsequent experiments. For cell cycle arrest which can prevent the spontaneous chromosome movement during experiments, the nocodazole treatment was carried out to prevent. As a first step towards the spatial control of chromosomes, the applied magnetics force was analyzed. To generate strong magnetic fields, the several neodymium magnets in various experimental condition were used. It is calculated that a large force of 780 fN was exerted on single BMP by a strong magnetic field. Based on the specific targeting test, we attempted to rotate cell using a remote magnetic field. The orientation of cell is dictated and arranged by the direction of a magnetic field. Next, we attempted to move the chromosomes located around the center of an oocyte by an external magnetic field. Without the antibodies, the BMPs did not conjugate with the chromosomes and the chromosomes did not move together with the BMPs in the presence of the magnetic field. The green spots from the FITC-conjugated BMPs moved in the direction of the magnetic field, whereas the blue spots from the DAPI-stained chromosomes remained close to the center. On the other hand, with antibody, the blue spots (chromosomes) coincided with the green spots (H1-BMPs) that noticeably moved to the top. For a more detailed analysis, chromosomes movement was quantitatively assessed by using image process. The mass center of the blue spots was calculated and used as a value that represents the entire distance moved. We compare the distances with and without. A substantial difference was observed between the cases with (20.59 ± 2.47 μm) and without (9.18 ± 4.23 μm) antibodies. To gain further evidence for chromosome movement by a remote magnetic field, we observed the time course motion of the chromosomes in live oocyte. To observe lateral movement, we prevented the rotation and drift of the living oocyte by anchoring it to the substrate. To achieve this goal, we developed a vacuum-assisted microfluidic device for trapping the oocyte on a transparent substrate and tested it. The oocytes were Hoechst-stainied before immobilized on the trapping hole. When the negative pressure was applied to the microfluidic device, oocytes were rapidly attracted to and fixed on the holes. A magnetic field was applied from the side of the trapped oocyte. The sequence of captured images shows the lateral motion of the chromosomes toward the applied magnetic field. The velocity of chromosome movement by a remote magnetic force was calculated as ~70 nm/min. Our data shows that the movement of the chromosomes linked to the H1-BMPs can be severely hindered by the meshwork of actin filament bundles. Recently, although human stem cell was generated by somatic cell nuclear transfer (SCNT), the enucleation process is still important factor in SCNT. It is well known that the invasive enucleation (removal of human oocyte genome) leads to the failure of embryonic development after genome exchange due to the loss of meiosis-specific factors associated with the spindle removal during physical enucleation process. Thus, we suggest the non-invasive method for SCNT without enucleation, by targeting chromosome with H1-BMPs. To achieve it, two possibilities have to be pre-verified. First, there are no interaction between chromosomes in somatic cell and unbounded H1-BMPs in oocyte after somatic cell – oocyte fusion. In previous work, we showed the possibility to control the both H1-BMPs and chromosome attached to H1-BMPs by using external magnetic field. Through previous result, the position of unbounded H1-BMPs can be controlled before somatic cell chromosomes are fused with oocyte chromosomes. Second, to deactivate chromosome activity, specific time (10 hrs) is necessary to target chromosome with H1-BMPs. Thus, it is required to investigate whether oocyte move to blastocyst stage after delay in activation for 10 hrs. Thus, it is investigated whether oocyte move to blastocyst stage after delay of activation for 10 hrs. It is shown that oocytes are well developed and reached the blastocyst stage with ~ 20 % despite of delay of activation time. Second there are no interaction between chromosomes in somatic cell and unbounded H1-BMPs in oocyte after somatic cell – oocyte fusion. These results show the possibility to apply our method to SCNT process. In the study described above, we demonstrate the possibility to non-invasively regulate a chromosome activity by the magnetic labeling, leading to arrest of embryonic development in parthenogenesis due to abnormal transcriptional activity. Through result, our non-invasive technique can be a candidate of alternative methods in somatic cell nuclear transfer (SCNT) that suffers from cell damage during enucleation process. To validate the potential application, the long range displacement of chromosomes is verified in oocyte by external magnetic field. We showed the traction of whole chromosomes in an oocyte across the entire cell span using functionalized magnetic nanoparticles (H1-BMPs). Procedures and techniques were well developed. In addition, our technique will provide a useful tool for investigating cellular functions associated with the spatiotemporal distribution of such components.
도시공간은 사람들이 밀집되어 있는 동시에 공간과 사람, 사람과 사람의 관계 등 상호 작용이 집약적으로 일어나는 대표적 공간이다. 우리는 일상생활을 통해 도시공간 속 여러 관계들을 포착함으로써 공간을 경험하고 심리적으로 체험하게 되며, 동시에 공간은 도시를 구성하며 사회적 영향 관계 속에서 의미를 획득한다. 즉, 도시공간 속 변화하는 인간의 심리적 체험은 건축 공간에 끼치는 영향을 분석하기 위한 중요한 점으로 위치하며, 기존의 물질적 토대를 바탕으로 하는 관계의 조직에 새로운 시각을 제시하는 차원에서 의미를 얻는다. 현대 도시공간의 특성과 소비적 심리를 반영하는 호텔은 도시에서부터 건축, 건축에서부터 내부공간, 내부공간에서부터 다시 도시로 향하는 이용객들의 경험과 심리적 체험을 독특하고 다양한 디자인으로 표현하며 새로운 소비계층들에게 각광받고 있는 장소이다. 이에 본 연구에서는 도시공간을 바라보고 건축 공간에 대한 사회ㆍ심리적 차원에서의 인식의 틀을 탐구하여 호텔에 대입해봄으로써 심리체험 기반 도심호텔의 새로운 유형을 제시하고자 한다. 건축 공간 개념으로서의 심리는 이용객의 공간을 점유하는 시간과 공간의 이미지 및 사회적 관계 형성에 영향을 미치며, 도시공간 속 살아가는 사람들의 공간적 체험을 가능하게 하는 중요한 변수가 되고 있는 것이다 Urban space is a representative space that shows the interactions and interrelationships between individuals and space. We learn and have a psychological experience from place by capturing various relationships in urban space through daily life. At the same time, the space constitutes the city and acquires the meaning through social interactions. In other words, the changing psychological experience of human beings in urban space is an important point for analyzing the effect on the architectural space, and it is gained in the meaning of presenting a new perspective to the organization of relationships based on the existing material foundation. Hotel where reflects the characteristics and consumerism of modern urban spaces expresses the physical and psychological experiences of users from city to architecture, architecture to interior space and interior space to the city again as a unique and diverse design. It is a place where is taken center stage by new consumer classes. In this study, we propose a new type of psychological experience-based urban hotel through applying research of social and psychological perceptions of the architectural space by view of the city to the hotel. Architectural psychology affects the image of time and physical environment and also the formation of social relationships. And it is an important variable that enables the spatial experience of people living in urban space.
공간을 아카이브하기 위한 시각적 표현 연구 : 문화재생공간 시흥시 맑은물상상누리를 중심으로
공간은 다양한 분야에서 다양한 관점에 의해 표현이 되고 있다. 본 논문은 그래픽디자인에서 공간을 어떻게 주체적으로 기록하고, 표현할 수 있을지에 대한 의문에서 시작되었다. 최근 그래픽디자인계에서 디자이너가 주체성을 가지고 저자성(authorship) 있는 작업의 한 방식으로 아카이브를 도구로써 활용하고 있다. 개념 미술의 아카이브 아트에서는 작가가 주체적으로 기록을 수집·분류·재구성하는 과정을 거치며 일련의 소재로부터 다른 의미와 형식을 만들고 있다. 그래픽디자인에서도 비슷한 맥락으로 디자이너가 주체성을 갖고 아카이브의 과정을 거치며 수집한 기록물을 재맥락화하는 작업이 유행하고 있다. 이러한 흐름은 그래픽디자인에서 아카이브가 하나의 장르가 되어가고 있으며, 아카이브는 기록학을 넘어 하나의 소통의 도구가 될 수 있음을 보여주고 있다. 아카이브 행위는 무심코 지나친 자료에 힘을 실어주는 역할을 한다. 이에 따라, 연구자는 타자에 의해 다양한 관점에서 표현되어 온 하나의 공간을 단순한 수치와 위치 등의 도면 정보로만 표현하지 않고, 고유한 관점을 통해 공간을 둘러싼 구조와 관계를 그래픽디자인에서 해석하고 기록하며, 시선의 위계(hierarchy)에 따라 정보를 재맥락화하는 방식을 연구하고자 하였다. 이 연구는 아카이브가 그래픽디자인에서 디자이너가 공간을 표현하는 또 하나의 도구가 될 수 있음을 증명하는 데 목적이 있다. 연구자는 그래픽디자인에서의 아카이브에서 디자인 프로세스를 문제인식(problem recognition)-취합(collect)-기록(document)-재구성(restructure)-맵핑(mapping)-그래픽디자인 콘텐츠로 전환(convert to graphic design content)로 정의하였다. 그중 정보를 맵핑하는 과정에서 목록(index) 구조를 형성하여 콘텐츠화할 수 있는데 이는 '취합(collect)-기록(document)-재구성(restructure)'의 과정을 거치는 아카이브 아트와 다른 점이라 판단하였다. 이러한 차이점은 개념 미술에서의 아카이브 뿐만 아니라, 건축에서의 아카이브와도 구별된다. 공간의 정보를 재구성하는 '목록(index) 구조'는 디자이너의 주체성과 의미를 생산하는 창작의 도구가 될 수 있음을 발견하고, 사례연구를 통해 공간을 기록한 디자인 작업이 어떠한 목록 구조로 구성되어 관점을 강화시킬 수 있는지 살펴보고자 하였다. 공간을 아카이브하기 위한 목록 구조는 공간을 바라보는 특정 시점, 다중 시점으로 나눌 수 있으며, 객관적 정보의 나열, 특정한 공간의 부분을 보여주는 해석적 정보의 나열이 될 수 있음을 파악하였다. 그래픽디자인에서의 아카이브 프로세스를 적용하기 위해 기반연구에서 공간을 아카이브하기 위한 연구대상지를 시흥시에 위치한 과거 정화처리장이었던 문화재생공간 맑은물상상누리로 선정하여, 공간이 재생되는 과정에서 역할이 변해가는 과정을 기록하고자 하였다. 본 연구대상지는 1989년부터 2024년 5월까지 정화처리장에서 유휴화된 공간, 부분적으로 개방된 공간, 전면 개방되어 문화재생공간으로 역할이 변화한 곳이기 때문이다. 1989년부터 다양한 기록자에 의해 수집된 자료와 연구자가 직접 수집한 자료를 취합하여, 사물·사람·공간으로 기록을 분류하고, 같은 대상과 위치를 과거와 현재에서 살펴볼 수 있는 목록 구조를 제작하고자 하였다. '있기 전과 후'에 대한 정보를 짝지어 재배치하기 위해 '있기에 앞서(Be, Fore)'라는 연구자의 해석점을 통해 목록의 주제를 설정하고, 기록물을 분류하고 목차에 반영하였다. 사물의 용도 변화, 사람들의 경험, 건축물의 변화 등에 대한 정보를 ‘있기 전과 후’의 구조로 재배치하였다. 이러한 기록물은 기록 시점, 기록자, 위치, 매체 등에 대한 정보를 담고 있다. 최종 작품은 기반연구에서 목록에 의해 재배치한 기록물을 출판물과 온라인 아카이브, 포스터의 형태로 제작하고자 하였다. 출판물은 목차를 이루는 각 장을 한 권씩 분리하여, 총 6권의 책과 1권의 부록으로 구성된다. 목차는 '1장 파편이 있기에 앞서', '2장 기억이 있기에 앞서', '3장 공간 개방에 앞서', '4장 공간의 개방', '5장 사람들의 기억', '6장 공간의 파편'이다. 책 구조는 각 장이 1권씩 분권 되어, 각 목록마다 기록의 전과 후를 짝지어 살펴볼 수 있게 구성하였다. 폐쇄된 시점과 개방된 시점을 표현하는 시각적 톤은 컬러, 이미지의 표현, 종이 등으로 시각적 대비감을 주고자 하였다. 목록 구조를 드러내기 위해 표지에는 기록물의 목록을 나열하였고, 기록물이 갖는 일정한 시선의 흐름을 위해 일정한 위치에 선을 배치하여 시간순에 의한 정보의 흐름을 유도하고자 하였다. 연구자의 해석적 표현에 치우치지 않기 위해 부록에는 공간에 대한 객관적 설명이 포함되어 있다. 온라인 아카이브에서는 목록화된 정보의 구조를 사용자에 의해 비선형으로 볼 수 있게 구성하였다. 온라인 아카이브는 인덱스, 맵을 통해 목록화된 정보를 비선형적으로 탐색하는 방법을 제안한다. 기록물의 전과 후를 비교할 수 있게 기록물을 연결하였으며, 맵 메뉴를 통해 시기별로 다양한 사람들이 기억하는 위치별 기록을 살펴볼 수 있게 구성하였다. 온라인 아카이브는 공간의 기록물을 어디서든 살펴보기 유용한 도구로서 대중의 접근이 쉽고, 지속적 업데이트가 가능하기 때문에 해당 공간의 기록물 콘텐츠를 담은 플랫폼의 역할을 기대할 수 있다. 포스터는 전시에서 아카이브 목록을 개괄적으로 살펴볼 수 있게 하는 역할을 한다. 최종 작품연구를 통해, 과거 오염된 하수를 처리하던 폐쇄적 산업시설이었던 정화처리장이 개방적 문화재생공간으로 변화하는 과정에서 비롯된 다양한 사람들의 기록이 모여 공간의 의미를 재조명할 수 있었다. 또한 공간에 관한 과거 기억과 현재를 파악할 수 있는 아카이브의 목록 구조를 형성함으로써, 그래픽디자인에서 공간을 아카이브한다는 것의 디자인적 가치를 확인하였다. 그러므로 본 연구는, 미시적으로는 '그래픽디자인에서의 아카이브 프로세스'를 통해 아카이브를 시각화한 맑은물상상누리 공간뿐 아니라, 또 다른 재생공간, 역사적 공간 등 과거의 흔적이 있는 곳에 적용될 수 있다. 거시적으로는 그래픽디자이너가 공간의 기록을 주체적으로 수집하고 정리하는 과정에서 디자이너의 관점이 담긴 목록의 위계가 형성되고 기록이 재배치됨으로써, 그래픽디자인에서 아카이브가 장르화되는 현상을 뛰어넘어, 공간의 의미를 재생산하는 창작을 위한 도구로서의 가능성이 있다는 점을 제시할 수 있을 것이다. Space is expressed from various perspectives across multiple disciplines. This study began with the question of how space can be subjectively recorded and represented within graphic design. Recently, in the field of graphic design, designers have been utilizing archives as a tool for authorial work, emphasizing their subjecthood. In archival art within conceptual art, artists create new meanings and forms through the subjective processes of collecting, classifying, and reorganizing materials. Similarly, in graphic design, there is a trend where designers recontextualize collected records through archival processes, emphasizing their subjecthood. This approach signifies that archives are becoming a genre within graphic design and a tool for communication beyond mere documentation. The act of archiving empowers materials that might otherwise be overlooked. Consequently, this study seeks to explore ways to interpret and record the structures and relationships surrounding a space in graphic design, rather than representing it solely through quantitative and locational data on diagrams. By examining and recontextualizing information based on the hierarchical viewpoint, the study intends to demonstrate that archives can serve as an additional tool for designers to express space in graphic design. The researcher defines the archival process in graphic design as problem recognition, collection, documentation, restructuring, mapping, and conversion to graphic design content. Among these, the process of mapping information involves forming an index structure to create content, which is identified as a key difference from the archival art process, which follows the stages of collecting, documenting, and restructuring. This distinction sets it apart not only from archival practices in conceptual art but also from those in architecture. The "index structure" for reconstructing spatial information can become a creative tool for generating the designer's subjectivity and meaning. This study aims to examine how design work documenting space can be structured using various index structures to enhance perspectives through case studies. The index structures for archiving space can be categorized into specific viewpoints and multiple viewpoints, presenting either a list of objective information or interpretative information showing specific parts of the space. To apply the archival process in graphic design, the foundational study selected MMSS(meaning "Clean Water Imagination Center"), a cultural regeneration space in Siheung City that was formerly a purification plant, as the study site. The goal was to document the changing role of the space throughout its regeneration process. From 1989 to May 2024, this site transitioned from a purification plant to an unused space, a partially opened space, and finally, a fully open cultural regeneration space. The study compiled data collected by various recorders since 1989, along with materials gathered by the researcher. These records were classified into objects, people, and space, and an index structure was created to compare the same subjects and locations in both past and present contexts. To rearrange information about "before and after" states, the researcher established the theme "Be, Fore" as an interpretative point, setting the topic for the lists and categorizing the records accordingly. Information on changes in object usage, people's experiences, and architectural transformations was restructured into a "before and after" format. These records include details on the time of recording, the recorder, the location, and the medium. The project aimed to produce the restructured records, as organized in the foundational research, in the form of both a book and a website. The book is divided into six volumes and one appendix, with each chapter separated into individual volumes. The chapters are titled: "Chapter 1: Before the Fragment," "Chapter 2: Before the Memory," "Chapter 3: Before the Opening of Space," "Chapter 4: The Opening of Space," "Chapter 5: People's Memories," and "Chapter 6: The Fragment of Space." Each chapter pairs records from "before" and "after" for comparison. The visual tones representing closed and open states are distinguished through the use of color, imagery, and paper types to provide a visual contrast. The cover features a list of the records to highlight the index structure, with lines placed at specific positions to guide the chronological flow of information. The appendix includes objective descriptions of the space to avoid skewing the interpretation towards the researcher's perspective. The website is designed to allow users to view the structured information non-linearly. The digital archive offers methods for exploring indexed information non-linearly through indices and maps. It connects records to compare the "before" and "after" states and allows users to examine location-specific records from different periods via the map menu. As a useful tool for viewing spatial records from anywhere, the digital archive facilitates easy public access and continuous updates, thereby serving as a platform containing the recorded content of the space. The poster serves to provide an overview of the archival index at the exhibition. Through the final project, the transformation of the former sewage treatment plant—once a closed industrial facility that processed polluted wastewater—into an open cultural regeneration space was documented. This documentation brought together various people's records, recontextualizing the meaning of the space and forming an archival index structure that captures both past memories and the present state of the space. This process confirmed the design value of archiving space within graphic design. Therefore, this study demonstrates that the application of the 'archival process in graphic design' can be used to visualize not only the MMSS space but also other regenerated or historically significant spaces. On a broader scale, it suggests that hierarchies of lists reflecting the designer's perspective can be formed, and records can be rearranged through the subjective collection and organization of spatial records by graphic designers. This process transcends the phenomenon of archiving as a mere genre in graphic design, proposing that it can be a creative tool for reproducing the meaning of space.

피부미용 산업은 고객과의 밀접한 접촉과 감정노동이 수반되는 특성을 가지며, 이러한 환경은 피부관리사에게 높은 수준의 직무스트레스를 야기한다. 이에 본 연구는 피부관리사의 직무환경에서 발생하는 스트레스 요인을 규명하고, 이로 인해 발생하는 프리젠티즘과 직무성과 간의 관계를 살펴보고, 회복탄력성이 이를 완화하는 데 어떠한 역할을 하는지를 규명하고자 시도하였다. 본 연구는 피부관리사 320명을 대상으로 설문조사를 실시하였으며, 이 중 신뢰성이 결여된 13부를 제외한 307부를 최종 분석에 활용하였다. 연구에서 직무스트레스는 직무요구, 조직체계, 직무불안정, 보상부적절, 직무자율, 관계갈등, 직장문화, 물리환경 등 8개 요인으로 구성되었으며, 프리젠티즘은 단일 요인으로, 직무성과는 직무수행과정과 직무수행성과로 구분되었다. 또한, 회복탄력성은 긍정성, 사회성, 통제성의 3개 요인으로 측정되었다. 연구 결과, 직무스트레스는 프리젠티즘에 유의미한 정(+)의 영향을 미치며, 보상부적절, 직장문화, 조직체계, 직무요구, 관계갈등 스트레스가 프리젠티즘을 증가시키는 주요 요인으로 나타났다(p<.001). 반대로, 직무스트레스는 직무성과에 유의미한 부(-)의 영향을 미쳤으며, 직무수행과정과 직무수행성과 모두 스트레스 요인이 높을수록 저하되는 것으로 나타났다(p<.001). 이는 직무스트레스가 피부관리사의 직무 만족도와 업무 성과에 부정적 영향을 미친다는 점을 실증적으로 확인한 결과이다. 또한, 회복탄력성은 직무스트레스와 프리젠티즘 간의 관계, 그리고 직무스트레스와 직무성과 간의 관계에서 유의미한 조절적 역할을 발휘하였다. 긍정성은 직장문화 스트레스와 프리젠티즘 간의 관계를 조절하여 긍정성이 높은 경우 직장문화 스트레스가 프리젠티즘에 미치는 부정적 영향을 완화하였다. 사회성은 직무자율 스트레스와 프리젠티즘 간의 관계를 조절하였으며, 통제성은 조직체계와 직무요구 스트레스가 직무성과에 미치는 부정적 영향을 줄이는 데 기여하였다(p<.05). 이상의 결과는 피부관리사의 직무스트레스가 프리젠티즘 및 직무성과에 미치는 부정적 영향을 완화하기 위해 회복탄력성을 강화하는 전략적 접근이 필요함을 시사한다. 본 연구는 피부관리사의 직무스트레스 관리 및 회복탄력성 강화를 위한 체계적인 교육 프로그램 도입의 필요성을 강조하며, 구체적인 실행 방안으로 긍정성, 통제성, 사회성과 같은 심리적 자원을 강화할 수 있는 맞춤형 훈련 프로그램 개발을 제안한다. 또한, 피부관리사의 보상체계 개선, 조직 체계 강화, 스트레스 완화를 위한 지원 시스템 구축 등의 조직적 노력이 필요하다. 본 연구는 피부미용산업의 특수성을 반영하여 프리젠티즘 예방과 직무성과 향상을 위한 실질적이고 전략적인 관리 방안을 제시하였다는데 의의를 둘 수 있다. The skin care industry is characterized by close client interactions and a high level of emotional labor, often results in significant job stress for estheticians. This study aims to identify the stress factors in estheticians' job environments, examine the relationship between job stress, presenteeism, and job performance, and explore the moderating role of resilience in these relationships. A survey was conducted with 320 estheticians, resulting in 307 valid responses after excluding 13 unreliable ones. Job stress was categorized into eight dimensions: job demands, organizational systems, job instability, inadequate compensation, job autonomy, relational conflicts, workplace culture, and physical environment. Presenteeism was measured as a single factor, while job performance was assessed through two components: process performance and outcome performance. Resilience was evaluated using three dimensions: positivity, sociability, and controllability. The results revealed that job stress significantly increased presenteeism, with inadequate compensation, workplace culture, organizational systems, job demands, and relational conflicts identified as major contributors (p<.001). In contrast, job stress significantly decreased job performance, affecting both process and outcome performance (p<.001). These findings empirically demonstrate the adverse effects of job stress on estheticians' job satisfaction and performance. Furthermore, resilience played a significant moderating role in the relationships between job stress and presenteeism, as well as between job stress and job performance. Positivity moderated the relationship between workplace culture stress and presenteeism, mitigating the negative impact of workplace culture stress on presenteeism when positivity levels were high. Sociability moderated the relationship between job autonomy stress and presenteeism, while controllability reduced the negative effects of organizational systems stress and job demands stress on job performance (p<.05). These findings highlight the need for targeted interventions to enhance resilience in order to mitigate the detrimental effects of job stress among estheticians. Training programs designed to strengthen psychological resources, such as positivity, sociability, and controllability, are recommended. Furthermore, organizational efforts to improve compensation structures, support systems, and workplace culture are essential. By addressing the unique demands of the skin care industry, this study offers actionable insights for developing effective strategies to reduce presenteeism and enhance job performance.
한국 동요에 관한 연구 : 국민학교 4.5.6학년 음악교과서를 중심으로

어린이가 노래를 부른다는 것은 가장 자연스럽고 보편적인 표현방법이며, 동요를 부르게 하는 것은 미성숙한 단계에 있는 어린이들에게 선율과 가사를 통해 예술성을 경험시키고 어린이들의 정신세계 형성에 중요한 역할을 한다는 점에서 교육적으로 큰 의미가 있다. 이러한 공통점은 세계 여러 나라의 동요에서도 확연하게 나타난다. 그러나 세계각국은 그 나라만이 갖고 있는 문화적 전통과 시대적 변화과정의 차이로 인해 동요는 서로 다른 모습을 보인다. 특히 우리 나라의 동요는 여러 가지 점에서 중요한 의미를 갖는다. 동요는 어려운 현실속에서 민족의 한을 달래주는 기능을 수행하는 동시에 어린이들에게는 꿈과 희망을 심어주는 노래로 존재해 왔다. 따라서 우리 나라 동요는 꾸밈없고 순수한 것이어야 하며 상업성과 오락성을 가져서는 안된다. 이에 본 연구자는 시대의 변화에 따라 달라진 동요의 발달과정을 연구해 보고 현재 국민학교에서 사용하고 있는 4, 5, 6학년의 음악교과서내의 한국동요와 최근 애창되고 있는 KBS, MBC 창작동요곡의 비교분석을 통하여 동요의 변화, 동요교육의 방향과 창작방향에 대해 연구해 보았다. 1920년대부터 현재에 이르기까지 창작동요는 여러 가지 면에서 다양한 형태로 변화하였다. 우선 동요의 조성이 다양해졌고, 박자도 2/4박자와 4/4박자 위주의 동요에서 3/4, 6/8, 12/8, 9/8박자 등으로 다양해 졌다. 곡의 형식면에서는 7.5조 운율의 동요가 주를 이루던 것에 비해 현재는 다양한 형식의 곡들이 많이 생겼으며, 6도 이상 도약음과 syncopation의 사용도 보편화되고 음역 또한 넓어졌다. 가상의 내용 역시 자연을 소재로 한 곡이 많았던 것에 비해 이제는 생활속에서 얻을 수 있는 모든 것들이 동요 가사의 소재로 다양하게 사용되고 있다. 이와 같이 시대적, 문화적 변화에 따라 동요는 변화해왔고, 이에 맞추어 어린이들에 대한 동요지도와 교육도 변화해야 한다. 학교 음악교육에 있어서 교과서내의 동요곡은 교육목표와 교육과정에 따라 교육해야 하며 어린이들에게 동요에 대한 관심을 갖도록 유도하기 위해서는 동요창작자나 동요교육자 모두의 노력이 요구된다. 작사, 작곡, 교육의 방법을 한마디로 이야기할 수는 없으나 다양한 형태의 창작과 지도방법이 필요하다. 가장 중요한 것은 어린이들에게 동요에 대한 관심과 애정을 갖게 하는 것이며 생활 속의 동요로서 온 가족이 함께 하는 동요가 되어야 한다. 사회적인 면에서는 방송이 동요에게 미치는 영향이 크므로 방송관계자와 동요지도자들의 세심한 연구와 노력이 요구된다. The first aim of this thesis is to compare with children's songs of textbooks and creative children's songs and to study characteristics, influence, necessity, educational value, process of change of the Korean children's song. Secondly, through this study I'm trying to seek for the standard way of teaching, education and composing of Korean children's song. The results of this study can be explained as following. 1. Children's song represents their society's own language, culture and history. children's sang provides pleasant life, development of socialization, and opportunity of self-expression to children and it becomes a conerstone of every education. 2. To compare with tonality of children's songs in textbooks and creative children's songs, tonality of creative children's songs are more varied than children's songs in textbooks. It's more educational for children to teach various kinds of tonality, especially folk key songs. 3. Meter and rhythm of children's songs in textbooks still mainly composed by rhythmed verse such as seven-five key, four-three key and four-four key. Therefore composers of children's song should make effort to open out rhythm of modern sense. 4. According to disparity of age, every range of voice is very different. Therefore individual ability and grade should be considered. 5. Sixth chard added-note chord and syncopation should be used properly oat to make children regared children's song as something bore and not interesting. 6. Main materials of words of children's songs are still natural objects. But as society progresses, words of children's sang should be changed iota modern sense, moreover we have to select every material that can express children's sense, feeling and mind. To compose and educate children's song, at first, we have to make effort to understand children's mind and recognize which is the right way to teach. Especailly, people in mass-media should keep puritism, artistism of children's song oat commercailism.
